摘要
目的 研究氟尿嘧啶 ( 5 -Fu)联合重组人生长激素 (rhGH)对肾癌细胞株的作用 ,探讨提高肾癌化疗效果的方法。方法 取对数生长期的人肾癌细胞株 (GRC -1) ,分为空白对照、rhGH、5 -Fu、rhGH +5 -Fu四组进行体外培养 ,2 4小时后测定各增殖周期的细胞比例值等指标。结果 在空白对照组 ,G0 -G1 期的细胞数占 5 3 .4%,S期细胞数占 42 .1%。在rhGH组 ,G0 -G1 期细胞数降低 ( 4 1.1%) ,S期细胞增高 ( 5 2 .7%)。在 5 -Fu组 ,G0 -G1 期细胞数增高( 62 .3 %) ,S期细胞数降低 ( 3 7.5 %) ;在rhGH +5 -Fu组 ,G0 -G1 期细胞数进一步增加 ( 67.4%) ,S期细胞数进一步降低( 3 2 .5 %) ,与 5 -Fu组比较差异有显著性 (P <0 .0 5 )。结论 重组人生长激素在体外能增强氟尿嘧啶抗肾癌细胞增殖作用。
Objective To study the effect of 5-Fu combined with rhGH on human renal carcinoma cell line for improving the effect of the themotherapy.Methods GRC-1 cell line were respectivelycultured for 24 hour in the following 4 groups:the control group,the rhGH group, the 5-Fu group, the rhGH+5-Fu group. The number of different cells were determined by flow cytometry(FCM).Results The percent of G 0-G 1/S phase respectively were in the 4 groups:53.4%/42.1% in the control group,41.4%/52.7% in the rhGH group,62.3%/37.5% in the 5-Fu group,67.4%/32.5% in the rhGH+5-Fu group.There were a significant difference in the rates of G 0-G 1/S phase cells between the 5-Fu group and the rhGH+5-Fu group.Conclusion rhGH might induce differentiation of GRC-1 cell and strengthen the effect of Fluorouraci in inhlibiting GRC-1 cell proliperation. This may be benefit to the chemtherapy of renal cell carcinoma.
