摘要
目的探讨人生长激素在体内对人胰腺癌细胞增殖和凋亡的影响。方法建立裸鼠胰腺癌移植瘤皮下模型,分为对照组、顺铂(DDP)组、重组人生长激素(rhGH)组和DDP+rhGH组,通过流式细胞仪、免疫组化和TUNEL法检测移植瘤的细胞周期、核增殖抗原、凋亡细胞。结果 DDP组和DDP+rhGH组比对照组或rhGH组S期细胞、核增殖抗原明显降低(P<0.05),凋亡指数明显升高(P<0.05);而rhGH组与对照组各项指标比较无明显差异(P>0.05)。结论外源性人生长激素在体内对人胰腺癌细胞的增殖和凋亡无明显作用。
Objective To study the effects of human growth hormone on proliferation and apoptosis of human pancreatic carcinoma cells of human in vivo. Methods Nude mice were randomly divided into control group,cisplatin (DDP) group,recombinant human growth hormone (rhGH) group and DDP+rhGH group after human gastric cancer xenograft model of nude mice was successfully established. The cell cycle,caryon proliferation antigen (PCNA),apoptosis index (AI)of xenograft were investigated by flow cytometry,immuno-histochemistry,and terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate biotin nick end labeling (TUNEL) respctively. Results Cells in S phase,PCNA of gastric cancer xenograft obviously degraded,but AI obviously increased in DDP group and DDP+rhGH groups compared with control and rhGH groups (P〈0.05). Between rhGH group and control group,there was no difference in above indices. Conclusion Exogenous human growth hormone has no obvious effects on proliferation and apoptosis of human pancreatic cancer cells in vivo.
