熊果酸对人舌鳞癌细胞株TSCCa的抑制作用及其机制探讨 被引量:27

摘要 目的 :研究女贞叶有效成分熊果酸对TSCCa细胞增殖的影响 ,探讨其中可能的机制。方法 :用MTT法研究熊果酸对TSCCa增殖的影响 ;用原位杂交的方法检测熊果酸对TSCCa细胞浆中核转录因子mRNA原位表达的影响。结果 :熊果酸抑制TSCCa细胞增殖 ,对TSCCa细胞的半数生长的抑制剂量约为 1 2 .5 μmol·L-1 ,在 2 4h内表现为一定的量效关系 ;原位杂交显示熊果酸对TSCCa细胞的抑制作用与抑制核转录因子的原位表达有关。结论 :熊果酸可能通过抑制TSCCa细胞的增殖来发挥抗肿瘤作用 ,其作用机制可能与抑制细胞浆中核转录因子mRNA的表达有关。 Objective: To study the inhibitory effect and possible mechanisms of ursolic acid on the proliferation of human tongue squamous carcinoma cell line TSCCa. Methods: Effects of ursolic acid on the growth were eva luated by MTT assay,and in situ hybridization of nuclear factor kappa B P65(NF kappa B P65) was also done. Results: The IC 50 value of ursolic acid for TSCCa cells was found to be 12.5 μmol·L -1 around and the expression of NF kappa B P65 was obviously inhibited by 25 μmol·L -1 and 12.5 μmol·L -1 ursolic acid when the cell line was exposed to ursolic acid for 1 day. In 24 hours, the inhibitory effect was positively related to the dosage. In situ hybridization showed 25 μmol·L -1 and 12.5 μmol·L -1 ursolic acid could inhibit the expression of NF kappa B P65 mRNA obviously. Conclusion: Ursolic acid can markedly inhibit TSCCa cells in 24 h in vitro and the inhibition of NF kappa B P65 mRNA may be involved in the mechanism.
