摘要
采用10%SDS-PAGE方法分析JY97-1/blosky和JY97-2/blosky各10个株系100个单株500粒F4籽粒的高分子量麦谷蛋白亚基(HMW-GS)组成,结果表明:1.Glu-D1位点的5+10.5+12的遗传在JY97-1/blosky后代中符合一对相对基因的分离。2.JY97-2/blosky后代HMW-GS组成类型变异极其丰富。在Glu-A1.Glu-B1和Glu-D1三个位点上分别检测到2.9和6种不同的亚基类型,在Glu-A1位点上1亚基和null的频率分别为52.85%和47.15%;Glu-B1位点上的变异最丰富,出现20;7+8+9;6+8等七种新的等位基因类型;Glu-D1位点存在2+5+10;2+12,5+10;5+12等四种新的等位基因类型。最优亚基组合1,7+8,5+10的比例为11.84%。3.两个组合中Glu-B1b或及Glu-B1c的不完全表达率分别为2.07%和1.9%。
The composition of high molecular weight glutenin subunit (HMW-GS) of F4 seeds of two crosses (JY97-l/blosky and JY97-2/blosky) were analysis by 10% SDS-PAGE. The results showed that the Glu-D1 subunits (5 + 10,5+10+12.5 + 12) segregation proportion of F4 seeds was 7 : 2 : 7 in cross JY97-l/blosky and the HMW-GS composition types of JY97-2/blosky were extremely abundant. 2, 9 and 6 different HMW-GS types were separately found in Glu-A1, Glu-B1 and Glu-Dl. At the Glu-A1 locus, frequency of subunit 1 and null was 52. 85% and 47. 15%. Variations of HMW-GS at Glu-B1 were the most abundant, there were seven new allelic genes appeared, as subunits 20 and 7 + 8 + 9, 6 +8 etc. At the Glu-D1 locus, there were 2 + 5 + 10, 2 + 12, 5+10, 2 + 5+12 etc. four new allelic genes. The rate of the best of composition of high molecular weight glutenin subunit (1,7 + 8,5 + 12) was 11. 84%. The incomplete expression of rate of Glu-B1b or Glu-B1c was 2. 07% and 1. 9%.
