摘要
目的 探讨SR BI在OxLDL刺激人血单核细胞表达致炎细胞因子mRNA中的作用。方法 用原位杂交方法研究OxLDL、SR A的特异性抑制性配体 Fucoidin、17β Estrogen对人血单核细胞SR BImRNA表达的影响及其与MCP 1、PDGF、bFGF、IL 10mRNA表达的关系。结果 OxLDL(10～ 30mg·L-1)能剂量依赖性刺激SR BImRNA表达 (均P <0 .0 0 1) ,Fucoidin(5 0～ 2 5 0mg·L-1)能拮抗 2 0mg·L-1OxLDL的诱导效应 (均P <0 .0 0 1) ,同MCP 1、PDGF、bFGF、IL 10四种致炎细胞因子mRNA的表达相一致。 17β Estrogen(1× 10 -9mol·L-1～ 1× 10 -5mol·L-1)抑制上述致炎细胞因子mRNA表达 (均P <0 .0 0 1) ,但剂量依赖性提高SR BImRNA表达 (P <0 .0 0 1) ;在 1× 10 -5mol·L-1时 ,OxLDL(10～ 30mg·L-1)能对抗Fucoidin(2 5mg·L-1)而刺激上述致炎细胞因子mRNA表达 (均P <0 .0 0 1)。结论 SR
Objective To explore the function of scavenger receptor class B type I(SR-BI)in the human peripheral blood monocyte expression of proinflammatory cytokines mRNA induced by oxidized low density lipoprotein(OxLDL). Methods With the digoxenin-labeled oligonucleotide-probes in situ hybridization, we investigated the effects of OxLDL, fucoidin and 17β-estradiol on the primarily-cultured human peripheral blood monocyte mRNA expression of SR-BI and relationship between its expression and the mRNA expressions of MCP-1, bFGF, PDGF and IL-10. Results The mRNA expression of SR-BI was significantly increased in a dose-dependent manner after incubating with OxLDL (10～30 mg·L -1)(P<0.001) and fucoidin (50～250 mg·L -1) distinctly inhibited OxLDL(20 mg·L -1) from inducing the expression of SR-BImRNA, which was consistent with expression of the above proinflammatory cytokines mRNA(P<0.001). 17β-estradiol(1×10 -9mol·L -1～1×10 -5mol·L -1)inhibited the mRNA expression of above proinflammatory cytokines, but notably increased expression of SR-BImRNA in a dose-dependent manner(P<0.001);in the precondition of 17β-estradiol (1×10 -5mol·L -1), OxLDL(10～30 mg·L -1) significantly increased the mRNA expression of the above proinflammatory cytokines antagonizing the inhibitory effect of fucoidin(25 mg·L -1)in a dose-dependent manner(P<0.001). Conclusion SR-BI may not participate in the process which OxLDL stimulates human peripheral blood monocyte to give expression to proinflammatory cytokines mRNA.
