摘要
利用新的荧光试剂2,3-二甲醛基喹喔啉(QDCA),建立了两种抗生素头孢拉定和头孢氨苄的同步荧光鉴别分析法。在介质H3BO3-NaOH(pH=8.8)缓冲溶液中,以激发单色器的波长差(Δλ=90nm)进行同步扫描测定。头孢拉定和头孢氨苄两者同时测定的线性范围分别为0.1-28μg·mL-1和0.1-33μg·mL-1。该方法操作简便、结果准确可靠,适用于微量分析。
The objective of this paper is establishing a new synchronous fluorimetry to simultaneously identify two antibiotics,cephalexin and cefradine with a new fluorescent reagent quinoxaline-2,3-dicarboxaldehyde(QDCA) The measurement is carried out in a H_3BO_3-NaOH buffer solution(pH=8.8) with Δλ=90 nm for synchronous scanning,which gave peaks at 432.6 nm and 518.5 nm for the cephalexin and cefradine,respectively.The cefradine can be determined directly,the corresponding synchronous fluorescent signal of the cephalexin at 432.6 nm is slightly interfered with that of the cefradine and should be corrected.The method of correction has been provided in this paper.Many important factors,e.g.,reaction temperature,reaction time,stoichiometry of the fluorescent reactions,synchronous fluorescent spectra,conventional fluorescent spectra,selection of Δλ,effect of acidity and selection of the buffer solution are all reported.The linear ranges for the simultaneous determination of cephalexin and cefradine were found to be 0.1-28 μg/mL and 0.1-33 μg/mL respectively.This method is simple,sensitive,reliable and suitable for micro-amount analysis.
