摘要
以薄皮核桃茎尖为试材 ,对其分化生长过程中的褐化现象进行初步研究 ,经多次试验表明 ,在抑制褐化及保证茎尖正常分化生长时期 ,主要采取二步法 :①接种于 1 2MS +活性碳 2g培养基中 ,置于 5℃冰箱中黑暗培养 7d后取出 ;②转接于DKW +6 -BA 1 0mg L +IAA 0 0 1mg L +2 0 %硫代硫酸钠 5ml L培养基中置于 2 5℃ ,无光照环境下培养 ,每隔 1 5~ 2 0d转换培养基 ,此方法抑制褐化效果为佳 。
Experiment was aimed on studing the problem of browning control during the shoot tip culture of Pellicular Walnut The results showed that the best way of preventive methods for the browning of explanted tissue is that the explants of Walnut was cultured in the dark on DKW medium added with 6-BA 1 0 mg/L +IAA 0 01 mg/L +20% Na 2S 2O 3 5 ml/L at 25 oC for 15~20 d after dark culture on 1/2 MS medium added with activated carbon 2 g at 5 ℃ for 7 d, and then, transplant the subculture plantlet on the same medium at intervals of 15~20 d
