摘要
目的 研究雷公藤多甙 (TⅡ )对大鼠异种肝移植急性排斥的抑制作用。方法 以雌性仓鼠为肝脏供者 ,雄性近交系Wistar大鼠为受者 ,循改进的Kamada法行原位肝移植术。供、受者各30只随机配对 ,平均分为 2组 :对照组和TⅡ治疗组 (30mg·kg-1·d-1)。观察两组受者生存期 ;分别取两组术后 2h、72h及排斥死亡时受者的肝脏标本 ,行HE染色检查。结果 对照组平均生存期为(7.1± 0 .35 )d ,TⅡ组 (7.2± 0 .5 2 )d ,两组比较 ,差异无显著性 ;对照组及TⅡ组结膜充血的发生率分别为 10 0 %、31% ,两组差异有显著性 (P =0 .0 0 1) ;术后肝脏标本HE染色检测 :(1)术后 2h对照组和TⅡ组均未见排斥反应发生 ;(2 )术后 72h对照组出现排斥反应 ,且明显 ,而TⅡ组表现相对较轻 ;(3)受者 7d左右死亡时 ,对照组排斥反应剧烈 ,TⅡ组排斥反应也很强烈 ,但不如对照组严重。结论 单独应用TⅡ 30mg·kg-1·d-1可轻度抑制大鼠异种肝移植急性排斥反应 ,但并不能延长受者的生存期 ;术后结膜充血可作为该模型急性排斥反应的一种表现 ,TⅡ可缓解这种体征。
Objective To study the inhibitive effects of TⅡ on acute rejection in hamster-to-rat liver xenotransplantation. Methods Female Golden hamsters and inbred male Wistar rats were used as donors and recipients, respectively. Orthotopic liver transplantation was performed by Kamada's cuff technique with some modifications. Thirty recipients were divided into two groups at random: untreated control group and ( n =15) and TⅡ-treated group ( n =15). Survival time of recipients was observed. Liver specimens at 2?h, 72?h after transplantation and at death of the recipients postoperatively were collected, respectively, and examined by HE stain. Results The survival time of control and TⅡgroups were ( 7.1 ± 0.35 ) and ( 7.2 ± 0.52 ) days, respectively ( t = 0.087 , P = 0.931 ). The rates of conjunctival congestion of control and TⅡgroups were 100?% and 31?%, respectively with the difference being significant between two groups ( P =0.001). Histologically, rejections were not seen in both contral group and TⅡgroup at 2?h after operation; rejection was obvious in control group at 72?h after surgery, while mild in TⅡgroup; although rejections were severe in both groups at the death of the recipients, but it seemed slighter in TⅡgroup than in control group. Conclusion As monotherapy, 30?mg·kg -1 ·d -1 TⅡ could suppress the rejection of this model mildly but couldn't prolong recipients' survival. Conjunctival congestion could serve as a sign of rejection of this model and TⅡcould relieve it.
