细胞免疫在异种肝移植免疫排斥反应中作用的研究

摘要 目的观察细胞免疫在异种肝移植急性排斥反应中的作用,并初步探讨其分子机制。方法建立仓鼠到大鼠原位肝移植模型,运用切脾、环磷酸胺、环孢霉素A处理受体,观察其对生存时间影响,并运用免疫组织化学方法检测移植肝中淋巴细胞的浸润情况及穿孔素、Fas-L的表达。结果异种肝移植术后受体必须同时加用抑制细胞免疫抑制剂环孢霉素A方可延长其生存时间,免疫排斥反应发生时移植肝中可见大量的淋巴细胞浸润,及穿孔素和Fas-L的表达。结论细胞免疫参与了异种肝移植免疫排斥反应,其作用与穿孔素、Fas一L的表达密切相关。 Objective To study the role of cellular immunity in the rejection of hamster-to-rat liverxenograft. Methods Several immunosuppression measures were given to the recipients, and the survival time ofthe recipients was observed. The lymphocyte infiltration and the expression of perforin and Fas - L were observedby immunohistochemistry. Results The recipients' survival lime could not be prolonged unless foe cyclosporine Awas given to suppress the cellular immunity together with other treatment measures. In the acute rejection of liverxenografts the infiltrating lymphocytes including CD4, CD8 subsets and the expression Of penforin and Fas-L werefound. Conclusions Cellular immunity participates in the acute rejection of liver xenograft, and its mechanism isrelated to the expression of perforin and Fas-L.
