摘要 本研究采用高效液相色谱分析法两次纯化步骤,部分纯化人血浆内源性钠泵抑制物(ESPI),进而分析其生物学特性:1.抑制Na,K-ATP酶,2.抑制钠泵活性,3.竞争性地抑制~3H-巴因与红细胞上的受体或Na,K-ATP酶结合,4.具有“剂量-反应”抑制作用,5.其抑制作用具有可逆性,6.结合于Na,K-ATP酶的E_2构型。 In this study, a 2-step high pressure liquid chromatography was employed to purify partially the endogenous sodium pump inhibitor from human plasma. Some of its biological properties were delineated: 1. inhibition of Na, K-ATPase; 2. inhibition of sodium pump activity; 3. competitive inhibition of ~3H-ouabain binding to the receptors on erythrocytes or the Na, K-ATPase; 4. 'dose-response' inhibition; 5. reversible inhibition; and 6. binding to Na, K-ATPase in F_2 comformation.
