摘要
制备抗哇巴因多克隆抗体 ,探讨最佳的免疫周期及免疫剂量。应用哇巴因 (ouabain) -牛血清白蛋白 (BSA)结合物为抗原免疫家兔 ,每周从兔耳缘静脉采血 ,监测哇巴因特异抗体的产生 ;以0 .2 5mg/次、1mg/次、1.75mg/次、2 mg/次四种不同免疫剂量进行最佳免疫剂量的研究。 5个月后颈动脉取血 ,进行抗体效价、亲和力及特异性的检测。结果显示免疫间隔为 6周、免疫剂量为 1.75mg/次时可以获得高效价、高亲和力和高特异性的哇巴因抗血清。
This study described the preparation of polyclonal anti ouabain antibody.Rabbits were immunized with bovine serum albumin (BSA) ouabain conjugates in 4 different doses of 0 25mg,1 0mg,1.75mg,and 2mg once immunization.The effects of the period of immunization (2weeks, 4 weeks and 6 weeks)on the production of antibody were also had been studied.The titer of antibody was measured once a week.The results have showed that the anti ouabain antibody was of hight titer, high affinity and high specificity for ouabain when the dose of BSA ouabain conjugates we used to immunize rabbits was 1.75mg and the period of immunization was 6 weeks.
