摘要
目的 建立用制备型HPLC制备石衫碱甲 (HuperzineA)的方法。方法 用常规的方法提取出石杉总生物碱。总生物碱经HPLC直接进样 ,柱子以 1 0 μSiO2 为填料 ,型号为 2 0× 30 0mm(Microsorb -MVTM) ,以氯仿 -甲醇 -氨水 (84 0∶2 4∶1 .2 )为流动相 ,流速为 8ml/min ,检测波长为 31 2 .7nm。结果 每次进样 3g石杉总生物碱 ,可以得到将近 1 80mgHuperzineA ,纯度达到 >90 %的工艺要求 ,重复性良好。结论 本方法与常规的分离方法比较 ,HPLC法制备石杉碱甲具有操作简单 ,重复性好 ,周期短 。
Aim To establish a method using HPLC for production Huperzine A.Methods Extraction alkaloids of Huperziaceae herbs by routine method and directly production Huperzine A by HPLC.The assay was conducted on a SiO 2 10μ(20×300mm) column with chloroform-methonal-ammonia (840∶24∶1.2) as mobil phase at a flow rate of 8ml/min.The detection wavelength was 312.7nm.Results The 180mg Huperzine A was obtained by injection 3g assemble alkaloids of Huperziaceae every time and the purity was more than 90%.Its repetition property was better.Conclusion The method can be used in industrialization product of Huperzine A.Compared with routine method,it has more advantages such as operation easier,production cycle shorter,lower cost and so on.
