摘要
建立用高效液相色谱法 (HPLC)测定石杉科植物中石杉碱甲 (huperzineA)含量的方法。方法与结果 :按照常规的方法提取出石杉总生物碱 ,用HPLC对总碱直接进行分析。色谱条件如下 :SpherisorbC18分析柱 ,检测波长 2 31.3nm ;甲醇∶水 ( 85∶15,体积分数 )作为流动相 ,流速 1ml/min。外标法测定。该方法的线性范围是 0 .4 1~ 6.15mg/L ;平均加样回收率为99.72 %~ 10 0 .0 9%。相对标准差为 0 .33%~ 0 .4 5%。结论 :建立了用HPLC测定石杉科植物中石杉碱甲含量的方法 ,该方法快速、灵敏、准确。
Objective: To establish a HPLC method for rapid determination of huperzine A content in Huperziaceae. Methods and Results: Total alkaloids of Huperziaceae were extracted by routine method and used directly for the HPLC analysis. The samples were subjected to chromatography on a Spherisob C 18 column and eluted with aqueous methanol (85%) at a flow rate of 1 ml/min. The huperzine A content was determined with a UV/Vis detector at 231 nm and calibrated using external standards. The linearity range of the method was 0.41~6.15 mg/L, the average recovery rate of huperzine A was determined to be 99.72%~100.09% and average RSD was between 0.33%~0.45%. Conclusion: A fast and sensitive specific HPLC assay for determination of huperzine A in Huperziaceae has been developed. [
