摘要
目的 观察 5 -脂氧合酶抑制剂对黑色素瘤细胞产生金属蛋白酶的作用。方法 以Transwellchamber测定a375细胞的侵袭性 ;以免疫酶联吸附及明胶酶谱法测定a375细胞产生MMP 2、MMP 9的量。结果 在 5~ 2 0 μmol·L-1 浓度范围内 ,5 脂氧合酶抑制剂NDGA、AA86 1和MK886剂量依赖性地降低a375细胞对人工基底膜的侵袭性 ,并剂量依赖性地抑制a375细胞产生MMP 2、MMP 9。结论 5 脂氧合酶抑制剂能抑制a375细胞产生金属蛋白酶 。
Aim To study the effects of 5 lipoxygenase inhibitors on the production of matrix metalloproteinases of melanoma cells.Methods Invasiveness of a375 cells was detected with Transwell chamber.The production of matrix metalloproteinases (MMP 2,MMP 9) of a375 cells were measured by ELISA and gelatin zymogram.Results In the range from 5~20μmol·L -1 ,NDGA,AA861 and MK886 reduced invasiveness of a375 cells through Matrigel membrane in a dose dependent manner and significantly inhibited the productions of MMP 2 and MMP 9 of a375 cells.Conclusion 5lipoxygenase inhibitors could suppress production of matrix metalloproteinases of a375 cells and be able to inhibit the metastasis of melanoma cells.
