摘要
目的:研究5-脂氧合酶抑制剂对黑素瘤细胞a375黏附的作用。方法:体外培养人黑素瘤细胞a375,分别以环氧合酶抑制剂吲哚美辛,5-脂氧合酶抑制剂去甲二氢愈创木酸(NDGA)、AA861及MK886孵育2 h,以光密度法测定a375细胞对细胞外基质蛋白的黏附,以流式细胞仪测定a375细胞整合素的表达,以双抗体夹心法测定a375细胞核因子NF-kB的含量。结果:吲哚美辛对a375细胞与细胞外基质蛋白黏附、细胞整合素表达及细胞核因子NF-KB含量没有明显影响。NDGA、AA861及MK886剂量依赖性地抑制a375细胞对Ⅳ型胶原的黏附及NF-kB活化,显著降低细胞整合素β1表达量。5-脂氧合酶抑制剂作用浓度为20 μmol/L时,NDGA、AA861、MK886处理组细胞黏附率分别为对照组的(52.3±11.7)%、(48.7±9.9)%、(50.8±8.4)V0,整合素β1表达量分别为55.3%、54.9%、47.6%,NF-KB活化率分别为67.7%、72.6%、58.3%,与对照组比较均有显著性差异(P<0.05)。结论:5-脂氧合酶抑制剂通过抑制黑素瘤细胞NF-kB活化,降低整合素β1的表达量,抑制a375细胞对Ⅳ型胶原的黏附。
Objective:To study the effect of 5-lipoxygenase inhibitors on adhesion of melanoma cell a375. Methods:Human melanoma cell a375 was cultured in vitro,then they were incubated with cyclooxygenase inhibitor indomethacin and 5-lipoxygenase inhibitor NDGA.AA861 and MK886. Adhesion of a375 cell to extracellular matrix was determined by optical density assay. The expression of integrins on a375 cell was detected by flow cytometry. The level of NF-kB in a375 cell was measured by Sandwich ELISA. Results: Indomethacin didnot affect the adhesion of a375 cell to extracellular matrix,the expression of integrins and activation of NF-kB. 5-lipoxygenase inhibitors NDGA.AA861 and MK886 could inhibit the adhesion of a375 cell to collagen W ,suppress NF-kB activation in a dose-dependent way and reduce the expression of integrin (3, in a375 cell. Treated with 20 μmol/L of NDGA,AA861 or MK886,the adhesion rates of treated group vs control group were (52. 3 ± 11. 7)%, (48. 7±9. 9)%, and (50. 8 ±8. 4)% , respectively; the expression rates of integrin β1 were 55. 3%, 54. 9%, and 47. 6% .respectively; the activation rates of NF-KB were 67. 7%,72. 6%, and 58. 3% respectively. There was significant difference between treated group and control group(P<0. 05). Conclusion:5-lipoxygenase inhibitors can suppress the expression of adhesion molecule by reducing NF-KB activation of a375 cell so as to inhibit melanoma cell adhesion to extracellular matrix.
