摘要
目的 初步探讨甲状腺乳头状癌 (PTC)中 p5 3基因突变及其与 p2 1WAF1 /CIP1 蛋白定量表达的关系。 方法 采用 PCR- SSCP、DNA序列分析、免疫组织化学、图像分析等技术 ,检测 4 1例 PTC和 10例甲状腺滤泡性腺瘤标本组织中 p5 3基因突变情况和 p2 1蛋白染色状况。 结果 (1)伴 p5 3基因突变 ( 组 )和不伴突变 ( 组 ) PTC的原发肿瘤 (T)、淋巴结转移 (N)及远处转移 (M)差异无显著性 (P>0 .0 5 )。 (2 ) 、 和 之间 , 和 、 和 、 和 的平均积分光密度值 (D)差异具有显著性意义 (P<0 .0 1)。 结论 (1) PTC的演进和 p5 3基因突变与否无直接关系 ,p5 3蛋白的空间构象可能发挥关键作用。 (2 )在 PTC的发生、发展中存在依赖野生型 p5 3蛋白诱导P2 1WAF 1
Objective\ To explore p53\|gene mutation and its relation with quantitative expression of the p21\+\{WAF1/CIP1\} protein in papillary thyroid carcinoma(PTC).\ Methods\ Seventeen archive samples enrolled in group Ⅰ, tweenty liquer nitrogen PTC storage samples.\ P53 gene purified with polymerase chain reaction single strand comformational polymorphism(PCR\|SSCP) method.\ P21 expression was quantitatively by mean optical density(D) method.\ Results\ (1)There were no statistically significant differences between group Ⅱ(without p53 gene mutation) and group Ⅰ(with p53 gene mutation) PTC in T, N, M(P>0 05).\ (2)The difference of D among group Ⅰ, group Ⅱ and group Ⅲ was statistically significant(P<0 01).\ The D levels between group Ⅲ and group Ⅱ PTC, group Ⅱ and group Ⅰ ones, group Ⅲ and group Ⅰ ones were also significantly different(P<0 01).\ Conclusion\ (1)There is no direct relationship between the progression of PTC and the p53\|gene mutation.\ (2)The expression of p21\+\{WAF1/CIP1\} in PTC is induced by wild p53\|protein, which serve as a regulative route during carcinogenesis and development of PTC.
