摘要
目的 :研究重组人生长激素 (rhGH)联合顺铂、丝裂霉素诱导前胃癌细胞 (MFC)凋亡的作用及其可行性 .方法 :通过MTT法检测rhGH联合顺铂、丝裂霉素和单独使用顺铂、丝裂霉素对MFC的体外杀伤作用 ,以IC50 和杀伤率表示 ;采用流式细胞技术、DNA凝胶电泳、透射电镜等技术检测细胞凋亡情况 .结果 :体外杀伤实验结果显示 ,rhGH联合顺铂组、rhGH联合丝裂霉素组抑制率分别为 86 6 5 % ,6 9 33% ;单独应用顺铂组和单独应用丝裂霉素组抑制率分别为 4 6 7% ,4 2 6 8% ;rhGH联合顺铂组、rhGH联合丝裂霉素组IC50 分别为 10 0ng/mL ,12 5 μg/mL .rhGH联合顺铂比单独使用顺铂时 ,对前胃癌细胞具有更强的杀伤作用 ,P <0 0 1,有极显著差异 ;rhGH联合顺铂组、rhGH联合丝裂霉素组用流式细胞技术检测有明显凋亡峰 ;DNA凝胶电泳呈明显的凋亡梯形条带 ;电镜观察有细胞凋亡形态学特征性改变 .结论 :根据本实验结果可推测胃癌围手术期应用rhGH联合化疗药物顺铂、丝裂霉素治疗 ,有良好的协同作用 ,可有效地诱导胃癌细胞凋亡 。
Objective: To study the effect and feasibility of apoptosis induced by combined recombinant human growth hormone(rhGH)with CDDP or rhGH with MMC on cultured mouse forestomach carcinoma (MFC) cell. Methods: IC 50 value and kill rate of combined rhGH with CDDP or with MMC and single CDDP or single MMC on cultured MFC cell were detected by MTT method. Apoptotic cell was analyzed by flow cytometry (FCM) using Annexin III FITC-PI staining method, DNA agarose gel electrophoresis and electron-microscopic. Sub-G1 peak was also detected by FCM. Results: The combined rhGH with CDDP or MMC could inhibit the growth of MFC cell, inhibited rate was respectively 86 65%, 69 33%, inhibited rate of single CDDP and single MMC were respectively 46 7%,42 68%,and induced apoptosis of MFC cell. IC 50 value of combined rhGH with CDDP and combined rhGH with MMC were respectively 100?ng/mL, 125?μg/mL. The combined rhGH with CDDP or combined rhGH with MMC was stronger kill effect more than single CDDP or single MMC, P<0 01, it is super-discrepancy. Meanwhile, the DNA ladder-shaped strip of apoptosis was showed by agarose gel electrophoresis. The characteristic change of apoptosis morphology was discovered by electron-microscopic. Conclusion: To induce tumor cell apoptosis was an effective way treating tumor during pre-operation and pro-operation when to make use of the treatment combined rhGH with CDDP or MMC which can take more inhibition to gastric carcinoma cells than single CDDP and single MMC. According to this experiment, it is inferred that combined recombinant human growth hormone with CDDP or MMC induce apoptosis of MFC cell, and the effect be related to induce G1 and M period of MFC by rhGH, and to induce gastric carcinoma cell apoptosis was their main mechanism.
