益肾降浊冲剂治疗慢性肾衰竭的实验研究 被引量:12

摘要 目的 :探讨益肾降浊冲剂治疗慢性肾衰竭 (CRF)的疗效和机理。方法 :采用 5 /6肾切除大鼠CRF模型 ,随机分为模型组、大黄治疗组、益肾降浊冲剂大、小剂量治疗组 ,观察各组Scr、BUN、Ch、TG、MDA、SOD和肾脏常规病理及超微结构改变。结果 :大黄组和益肾降浊冲剂大、小剂量组肾脏病理均显著轻于模型组 ,而其中益肾降浊冲剂大、小剂量组病变最轻 (P <0 .0 1及P <0 .0 5 ) ,血浆Scr、BUN、Ch、MDA、SOD的结果同前。肾脏超微结构的改变与光镜下观测的肾脏病理结果相同 ,益肾降浊冲剂组的病变最轻 ,线粒体形态无明显改变。结论 :益肾降浊冲剂可以延缓CRF的进展 ,作用强于单味大黄 ;其机理可能是减少自由基 。 Objective:To study the effect and mechanism of Soluble Granules Yishenjiangzhuo Therapy on CRF. Methods:Forty Sprague-Dawley Rats (SD Rats) were randomly divided into five groups. Eight rats served as normal control group. 5/6 subtotal nephrectomy were performed in the other rats to establish the CRF model. One month after nephrectomy the CRF model rats were subdivided randomly into 4 groups: the model control group, the Chinese Rhubarb group, the large and small dose Soluble Granules Yishenjiangzhuo groups. Rats of the large and small dose groups received oral administration of Soluble Granules Yishenjiangzhuo in a dose of 20.0 kg/d and 10.0 kg/d for two months respectively and the rats of Chinese Rhubarb group were treated Granules of Chinese Rhubarb at a dose of 0.21 kg/d (equal to a dose of 0.5 kg/d raw material). Results:(1) Significant differences of BUN, Scr, MDA, SOD and the score of glomerulus/tubules pathology were observed between the model control group, Chinese Rhubarb group, the large and small dose Soluble Granules Yishenjiangzhuo groups ( P <0.01 or P <0.05), and between the Chinese Rhubarb group and the large/small dose Soluble Granules Yishenjiangzhuo group ( P <0.01 or P <0.05). (2) Changes of renal ultrastructures were the same as its pathology findings under light-microscope, damages of mitochondria was most serious, that of the Chinese Rhubarb is the secondly more serious, that of the large/small dose of Soluble Granules Yishenjiangzhuo is the slightest in all. Conclusion:Soluble Granules Yishenjiangzhuo can slow down the progress of CRF, and its effect is better than that of Chinese Rhubarb. Its mechanism may be due to the reducing of free radicle, protection and improvement the function of mitochondria.
