摘要
目的 探讨梭曼中毒对心脏胆碱能信号通路的影响。方法 利用RT PCR技术检测心肌乙酰胆碱能受体(M2 受体 )mRNA的表达 ,以蛋白印迹法 (Westernblottechnique)检测抑制性鸟苷酸结合蛋白 (Giα)含量 ,放射免疫法测定环磷腺苷 (cAMP)浓度及酶放射化学分析测定腺苷酸环化酶 (AC)、蛋白激酶A (PKA)活性。结果 与正常对照组相比 ,梭曼中毒大鼠心肌M2 受体mRNA的表达和Giα含量均明显降低 ,cAMP浓度及AC、PKA活性显著升高。
Objective To investigate the effects of soman intoxication on cholinergic signal transducting system in rat heart.Method The expression of acetyl-cholinergic receptor M 2 mRNA was detected by RT-PCR technique and concentration of inhibitory guanylic acid integrated protein(Giα)in the rat myocardium was measured with Western bolt technique.Concentration of cyclic adenosine monophosphate(cAMP)was measured with radioimmunoassay(RIA),activities of adenosine cyclase(AC)and protein kinase A(PKA)were measured by enzymic radiochemical methods with [γ- 32P]-ATP.Result The expression of M 2 receptor mRNA in the rat hearts was down-regulated significantly by soman intoxication,as compared with the controls(P<0.01),and dropped to the lowest six hours after intoxication.Giα concentration decreased by 12.2%(P<0.05),15.6%(P<0.01),and 20.6%(P<0.01)at the time of 6 hours,12 hours and 24 hours after intoxication,respectively,while concentration of cAMP and activities of AC and PKA increased markedly in the rats with soman intoxication.Conclusion Abnormal enhancement of cholinergic signal transducting system could play an important role in heart injury induced by soman intoxication in rats.
