摘要
以杂交狗牙根品种‘Tifeagle’的匍匐茎节作外植体 ,诱导出胚性愈伤组织 ,进一步分化后获得了再生植株。在再生植株中出现了体细胞变异 ,其中一株TV4的匍匐茎节间长度比亲本减短了 4 7% ,叶长减短了 32 % ,叶宽增大了 30 % ,根系较亲本更发达 。
The stolonous nodal segments of hybrid bermudagrass cultivar'Tifeagle'( Cynodon dactylon×C.transvaalensis )were sterilized by 0 1% HgCl 2 and placed on solid medium contained N 6 macronutrient elements,B 5 micronutrient elements,MS ferric salt, Gamborg's vitamin and additives including 2mg/L 2,4 D and 0 02mg/L 6 BA for induction of callus.The compact embryogenic calli were obtained and were highly regenerable after they were transferred to regeneration medium.Somaclonal variation was observed among the plantlets.One of them,TV4,showed dwarfism variation.Compared to its parent'Tifeagle',TV4 showed significant morphological variation with decrease of 47% in stolon internode length,increase of 30% in blade width,and reduction of 32% in blade length.TV4 had extensive root system with increase of 37% in dry weight of the newly developed adventitious roots after the stolon was planted for 40 days.TV4 showed higher chilling tolernace than'Tifeagle'with lower relative electrolyte leakage after the plants were subjected to 8℃.SOD activity decreased in'Tifeagle',while that increased in TV4 after chilling treatment.The activities of CAT and POD declined in both TV4 and'Tifeagle',but they were maintained at higher level in TV4 than'Tifeagle'.
