摘要 目的:探讨阿司匹林对结直肠癌化学预防的机理。方法:培养结肠癌细胞株Caco-2,用不同浓度阿司匹林进行干预,对细胞生长、增殖、凋亡及Fas/FasL表达进行观察、检测。结果:高浓度阿司匹林组细胞大量死亡,低浓度组促进细胞凋亡;25 mmol/L阿司匹林组可见Fas表达,而对照组不表达;25 mmol/L阿司匹林组FasL表达与对照组相比明显降低。结论:低浓度阿司匹林促进结肠癌细胞凋亡,其机理与诱导Fas表达、下调FasL表达有关。 Purpose: To study the mechanism of chemoprevention of colorectal cancer by aspirin. Methods: Different concentrations of aspirin have been used in the culture of colonic cell-Caco-2, its proliferation and apoptosis was observed during the culture and the expression of Fas/FasL mRNA was assayed by RT-PCR. Results: Major portion of cells died in the culture medium that was added high concentration of aspirin, but the lower concentration promoted apoptosis. The Fas mRNA was expressed in cells added with 25 mmol/L aspirin, but not in that of 1640. The FasL mRNA expressed in 25 mmol/L aspirin was lower than in 1640. Conclusions: The lower concentration of aspirin can promote apoptosis of colonic cell, the mechanism has correlation with induction of the expression of Fas, and down-regulation of the expression of FasL.
