在HBV感染的肝癌组织中Fas/FasL表达的作用

摘要 目的探讨Fas/FasL(Fas配体)在原发性肝细胞肝癌(HCC)组织中的表达及其与乙肝病毒(HBV)感染的关系。方法用免疫组化S-P法检测21例肝癌组织及其10例癌旁组织Fas/FasL的表达。结果在肝癌和癌旁组织中Fas表达的阳性率分别为52.38%和80.00%(P<0.05),FasL分别为66.67%和40.00%(P<0.05)。在HBsAg阳性和阴性组Fas表达的阳性率分别为50.00%和66.67%(P>0.05);FasL分别为61.11%和100%(P<0.05)。肝癌组织中Fas、FasL表达与性别、年龄、肿瘤大小无关,与癌栓转移呈负相关,FasL与分化程度有关。结论肝癌细胞能下调Fas及上调FasL的表达而使凋亡受阻,HBV感染能抑制肝癌组织FasL的表达,可能是HBV在HCC的发生及发展过程中的致癌机制。 Objective To explore the expression of Fas/FasL in hepatocellular carcinoma (HCC) and its relationship with HBV infection. Method The expressions of Fas/FasL were detected by immunohistochemistry (SP) from 21 cases of hepatocellular carcinoma as well as 10 corresponding non-cancerous tissues. Results The positive rate of Fas protein expression in hepatocellular carcinoma and corresponding non-cancerous tissues were 52.38 % and 80.00% respectively (P〈0. 005). FasL were 66.67% and 40.00% respectively (P〈0. 005). Whereas among HCC patients, the positive rates of Fas expression of HBsAg positive group and HBsAg negative group were 50.00% and 66.67% respectively (P〉0. 005). FasL were 61.11% and 100% respectively (P〈0. 005). The expression of Fas/ FasL have no correlation with sex,age and the size of tumor and have negative correlation with tumor embolus metastasis. The expressions of Fas were related with differentiation of hepatoma. Conclusions The Hepatoma cells can down-regulate the expression of Fas and up-regulated the expression of FasL to hinder apoptosis. The infection of HBV can inhibit the expression of FasL in hepatoma tissues, which may be the possible mechanism of carcinogenesis.
