摘要
目的 :探讨P2 1waf1、MDM2、P5 3蛋白在原发性肝细胞癌 (HCC)中的表达及其对于HCC诊断的意义。方法 :应用免疫组织化学SP法检测 6 1例原发性肝细胞癌及 5 9例相应癌旁肝组织中P2 1waf1、MDM2和P5 3蛋白的表达 ,并对它们之间的关系及其与肝癌的临床病理特征的关系进行统计学分析。结果 :P2 1waf1、MDM2和P5 3蛋白在HCC中的阳性表达率分别为 31.15 % (19/ 6 1)、2 6 .2 3% (16 / 6 1)和 5 7.38% (35 / 6 1) ,明显高于癌旁肝组织 (P <0 .0 0 1)。HCC中P2 1waf1与MDM、P5 3蛋白表达无明显相关性 (P >0 .0 5 )。这三种蛋白的表达与肝硬化及不典型增生无关 (P >0 .0 5 )。结论 :P2 1waf1、MDM2和P5 3高表达均在HCC的发生中起作用 ,但不是早期事件 ;HCC中存在不依赖wt-P5 3途径的P2 1waf1的表达 ,HCC中P2 1waf1的表达不能反映 p5 3的基因状态。
Objective:To investigate the expression of P21 waf1 ,MDM2 and P53 proteins in hepatocellular carcinoma(HCC) and its significance for HCC diagnosis.Methods:Using streptavidin-peroxidase conjugation method (SP), the expression of P21 waf1 ,MDM2 and P53 proteins was observed in 61 cases of HCC and 59 cases of corresponding paracancerous tissue. Statistic analysis was made to figure out the correlation between the proteins and the clinical pathological features of HCC.Results:Positive rates of P21 waf1 ,MDM2 and P53 immunostaining in HCCs were 31.15%(19/61), 26.23% (16/61) and 57.38% (35/61) respectively.The expression of the proteins in HCCs was significantly higher than that in paracancerous tissues ( P <0.001). The expression of P21 waf1 was not significantly correlated to that of P53 and MDM2 ( P >0.05). The expression of the proteins was not significantly correlated to cirrhosis and atypical hyperplasia.Conclusions:P21 waf1 、MDM2 and P53 overexpression takes part but is not early events in the pathogenesis of HCC;there is wt-p53 independent P21 waf1 expression in HCC and P21 waf1 expression in HCC can not reflect p53 gene status.
