摘要
目的为了在可注射组织工程骨支架材料中正确使用氯化钙,研究不同含量氯化钙对骨髓基质成骨细胞的细胞毒性作用。方法从兔髂骨中获取骨髓基质成骨细胞,将骨髓基质成骨细胞与不同含量氯化钙溶液进行不同时间的接触,然后再在细胞培养液中培养24h,观察骨髓基质成骨细胞的形态改变和细胞代谢活性的改变。结果骨髓基质成骨细胞与0.8mol氯化钙接触20min以上即可引起细胞代谢活性降低及引起细胞形态发生改变,0.8mol氯化钙在20min之内对成骨细胞的代谢活性没有明显影响,0.9mol氯化钙在15min之内对成骨细胞的代谢活性没有明显影响,1.0mol含量氯化钙在5min时间之内即对成骨细胞的代谢活性有明显影响。结论0.8mol氯化钙与骨髓基质成骨细胞接触20min之内无明显细胞毒性作用。大于0.8mol氯化钙与骨髓基质成骨细胞接触15min以上即有细胞毒性作用。
Aim To study the cytotoxicity of different levels of calcium chloride on osteoblast.Methods The osteoblasts were isolated from iliac bone of 3 New Zealand rabbits and then cultured in DMEM medium for 24 hours after they were contacted with different concentration of calcium chloride in different time.The osteoblasts were studied morphologically and metabolically.Results The osteoblasts contacted with 0.8 mol calcium chloride for more than 20 min caused their metabolism activity reduced and caused morphology changing,but within 20 min nothing changed significantly in the metabolism activity of osteoblasts.Within 15 min,0.9 mol calcium chloride showed no significant effect on the metabolism activity of osteoblasts,while the metabolism activity of osteoblasts significantly influenced by 1.0 mol calcium chloride within 5 min.Conclusion 0.8 mol calcium chloride contacted with osteoblasts for less than 20 min has no significant cytotoxicity.More than 0.8 mol calcium chloride contacted with osteoblasts for more than 15 min has the significant cytotoxicity.<P>
