摘要
利用PDS1000/氦气基因枪将人工合成的Bt基因导入玉米自交系340,8902,吉853,Mo17等的愈伤组织,在含有除草剂Basta2~5mg/L的培养基上筛选与分化。经3轮的除草剂筛选获得1840块抗性愈伤组织,再生860个植株。Southernblot分析证明,3个植株中整合有Bt基因。
Calli of maize inbreds 4112,7922,340,8902,Ji853,and Mo17 were bombarded using PDS 1000/He particle delivery system in the presence of plasmid pGBIL which contained a Bt gene and selectable marker Bar gene Resistant calli were seleted on Basta 2~5 mg/L containing medium 860 green plants were regenerated from 1 840 resistant calli Southernblot analysis of 3 green plants confirmed stable integration ofBt gene into Maize genome
