肉鸡腹水症发病机理研究:Ⅳ.低温处理对肉鸡心肌肌浆网钙泵活性与Ca^(2+)摄取的影响 被引量:2

摘要 采用超速离心法分离提纯右心室肌原纤维和肌浆网 ,测定低温处理早期肉鸡心肌肌浆网钙泵 (Ca2 + ATP酶 )活性和肌浆网Ca2 +离子浓度 ,以揭示低温处理早期肉鸡右心室功能增强和肉鸡肺动脉高压的机制。结果表明 :1) 19、2 7、33和 4 0d低温处理组SRCa2 + ATP酶活性较对照组显著性升高 ,33d时活性达到最高。肺动脉高压组、肺动脉低压组SRCa2 + ATP酶活性较对照组显著性升高 ,其中肺动脉高压组SRCa2 + ATP酶活性是低压组的 3倍 ,且差异极显著 (P <0 .0 1)。 2 )低温处理组 19和 2 1d心肌肌浆网Ca2 +含量明显升高 ,与对照组相比差异显著 (P <0 .0 5 )。33和 4 0d低温处理组肌浆网Ca2 +摄取趋向减少 ,与对照组相比 ,差异不显著。肺动脉高压组与对照组相比 ,肌浆网摄取Ca2 +的浓度显著性升高 ,差异显著 (P <0 .0 5 ) ;而且肺动脉高压组心肌肌浆网摄取Ca2 +的量显著性高于肺动脉低压组 (P <0 .0 5 )。 In this experiment, the ventricular myofibrillar and sarcoplasmic reticulum were purified from right ventricular by supercentrifuge. In order to illustrate the mechanism of enlargement in right ventricular, the activity of SR Ca 2+ ATP and Ca 2+ concentration were determined after exposed to low temperature. The results are as following: ①the activity of SR Ca 2+ ATP increases significantly as compared with the control group on day 19, 27, 33 and 40, particularly the activity amounts to the maximum on day 33. If the birds were divided into the pulmonary hypertension group, pulmonary hypotension group and control group, the activity of Ca 2+ ATP in pulmonary hypertension and hypotension group increased significantly compared with the control group. Furthermore, the activity of pulmonary hypertension group was three times of the pulmonary hypotension group(P<0.01). ②The concentration of SR Ca 2+ rose significantly on day 19 and day 21 after exposed to low temperature (P<0.05), but on day 33 and day 40, the concentration decreased, and there no significantly difference compared with the control group (P<0.05). The concentration of the pulmonary hypertension group was more than that of pulmonary hypotension group. The abore results suggest that the enlargement of ventricular function may have the relation to the activity of SR-Ca 2+ and the concentration of Ca 2+ in sarcoplasmic reticulum.
