细胞间隙连接蛋白在多种类型癌组织中的原位表达研究 被引量:16

摘要 背景与目的:恶性肿瘤细胞常有间隙连接蛋白(Connexin,Cx)的异常定位和表达,本研究目的是检测Cx基因家族中的Cx26、Cx32、Cx43和Cx45基因在多种类型癌组织微阵列中的原位表达,期望建立Cx26、Cx32、Cx43和Cx45基因在多种类型癌组织中的蛋白表达谱,探讨其在肿瘤发生、发展中的作用。方法:利用自制的组织微阵列制作仪制作含多种类型癌标本的组织微阵列,免疫组化法检测Cx26、Cx32、Cx43和Cx45基因在292例癌组织及89例癌旁组织中的表达。结果:(1)成功地制作出100点、200点和360点3种不同规格的组织微阵列蜡块和切片;(2)获得Cx26、Cx32、Cx43和Cx45基因在20种癌组织中的原位表达谱,Cx26、Cx32、Cx43和Cx45基因在肠癌、肝癌、胃癌、肺癌及甲状腺癌组织中的表达明显低于其相应的癌旁组织。结论:Cx26、Cx32、Cx43和Cx45基因的失表达对多种癌的发生有重要作用。多肿瘤的组织微阵列为肿瘤基因表达谱如Cx基因的研究提供一种高通量工具。 BACKGROUND &OBJECTIVE:Malig nant cells are often associated with aberrant expressio n or localization of connexins.Expr ession of Cx26,Cx32,Cx43,and Cx45g enes were studied to g ain their expression profile in tissue microarrayers con sisting of various carcinomas and to elucidate their function in carcinog enesis.METHODS :Basing on the principle of constructing tissue microarrays,t he authors take use of self-made tiss ue arrayer to construct tissue microarrays for the different kinds of cancer specim ens.Meanwhile,expression of connexin(Cx )g enes,such as Cx32,Cx26,Cx43,and Cx45in the tissue microarrays we re detected in 292cases of tumor tiss ues and 89paratumor tissues by immunohi stochemistry.RESULTS :(1)Three kinds of tissue microarrays consisting sa mples from 20different carcinomas w ere constructed successfully,which contained 360,200,or 100spots in each receptive paraffin block with tissu e cylinder of 0.75mm or 0.6mm diamete r respectively.(2)The expression profiles of Cx32,Cx26,Cx43,and Cx45in 20kinds of carcinomas and their relate d adjacent-cancer tissues were g ain ed,which were widely low expressed in ca rcinomas especially in nasopharyng eal carcinomas.Expression of Cx26,Cx32,Cx43,and Cx45in carcinomas such a s carcinomas of colon and recta,hepat ocellular carcinomas,g astric carc inomas ,lung carcinomas and thyroid cancers were sig nificantly lower than their related adjacent-cancer tissues.CONCLUSION:Low expression of Cx26,Cx32,Cx43,and Cx45mig ht play crucial roles in c arcinog enesis of many kinds of carcinomas.Tissue microarrays con sisting of many different kinds of ca rcinomas provide a new hig h-throug hout tool f or rapid and comprehensive molecula r expression profiles of carcinomas ,such as connexin g enes.
