摘要
目的了解细胞间隙连接蛋白43(Cx43)和上皮-钙黏蛋白(E-cad)在胰腺癌组织中的表达及其与胰腺癌发生和发展的关系。方法应用免疫组织化学PV6000法检测Cx43和E-cad在47例胰腺癌和10例正常胰腺组织中的表达和定位,并进行相关性分析。结果Cx43和E-cad在正常胰腺组织导管上皮细胞及腺泡细胞均呈阳性表达,而胰腺癌组织中Cx43和E-cad表达阳性率分别为44.7%(21/47)和46.8%(22/47),二者间差异有显著性(χ2=18.59、19.23,P<0.01)。Cx43和E-cad在组织学Ⅰ级的胰腺癌组织中的阳性表达率(70.0%、80.0%)明显高于组织学Ⅲ级(28.6%、23.8%),差异有显著性(P=0.047、0.021)。Cx43和E-cad阳性表达率与胰腺癌的转移密切相关(χ2=6.01、4.70,P<0.05),与病人1年生存率无关(χ2=2.07、3.63,P>0.05)。Cx43和E-cad在胰腺癌组织的表达呈显著正相关(χ2=16.31,r=0.92,P<0.01)。结论胰腺癌存在Cx43和E-cad的表达下调和定位异常,其可能是胰腺癌发生和发展过程中的重要环节。
Objective To study the expressions of connexin43 (Cx43), and E-cadherin (E cad) in the tissue of human pancreatic cancer and to study their relationship with the carcinogenesis of the disease. Methods By using immunohistochemical PV 6000 method, the expressions of Cx43 and E-cad in 47 cases of pancreatic cancer and 10 normal pancreases were detected and analysed. Results Expressions of Cx43 and E-cad were found in ductal and acinar cells in all the normal pancreatic tissues (100%). The positive rates of Cx43 and E-cad in the cancer were 44.7% and 46.8 %, respectively. The differences between them were significant (χ^2=18.59,19.23;P〈0.01). The positive rates of Cx43 and E-cad in grade- 1 were higher than that in grade Ⅲ (P= 0. 047,0. 021). Their positive expressions were closely associated with metastasis (χ^2=6.01,4.70;P〈0. 05), but not with the one-year survival (χ^2=2.07,3.63;P〈0.05). There was a positive correlation between the expressions of Cx43 and E-cad in the disease. Conclusion Down-regulation in the expression level of Cx43 and E-cad exists in pancreatic cancer, which might be an important link in the occurrence and development of this malignancy.
