摘要
①目的 探讨SEB对NK细胞杀瘤活性的影响。②方法 采用补体裂解法分离富集NK细胞,分别用10^(-9)g/ml、10^(-8)g/ml、10^(-7)g/ml、10^(-6)g/ml浓度的SEB作用NK细胞,分别于12、24、36、48h后测定NK细胞对K562细胞的杀伤活性。③结果 SEB处理NK细胞12h后杀瘤活性增强,至24h杀瘤活性最佳;且在一定浓度范围内NK细胞的杀瘤活性随SEB浓度的升高而增强。④结论起抗原SEB在体外能够增强NK细胞的杀瘤活性。
Objective To get a new way that adopted to tumor immunotherapy by studying the tumor - cytotoxic effect of NK cells induced by the super - antigen SEB. Methods Using complement lysis to rich NK cells; NK cells were induced by SEB of different concentrations: 10-9g/ml.10-8g/ml.10-7g/ml.10-6g/ml. The tumor -cytotoxic effect of NK cells was determined by MTT after being treated by SEB 12h,24h,36h and 48h. Results The tumor - cytotoxic effect was enhanced after being induced by SEB 12h later,and the optimal time is 24h. The effect was higher as the concentration of SEB increasing. Conclusion Super - antigen SEB can enhance the tumor - cytotoxic effect of NK cells in vitro.
