摘要
目的 探讨血小板对外周血纯化、扩增的NK细胞杀伤作用的影响.方法 使血小板与K562细胞在体外共孵育,以相差显微镜和流氏细胞仪检测血小板与K562细胞的粘附.从外周血单个核细胞(PBMC)中分离纯化、扩增NK细胞,以MTT法研究血小板对NK细胞杀伤活性的影响.结果 K562细胞体外能结合血小板,粘附率达到(25.0±4.3)%.血小板与K562细胞的粘附可抑制NK细胞对K562细胞的杀伤作用,且血小板对NK细胞杀伤活性的抑制呈数量依赖性.结论 血小板能粘附肿瘤细胞并能抑制NK细胞对K562细胞的杀伤作用.
Objective: To observe the inhibition effect of platelet on the lysis action of K562 cells by natural killer (NK) cells purified and proliferated from peripheral blood mononuclear cells(PBMC). Method: K562 cells were co-incubated with fresh isolated platelets in vitro, The adhesion of platelets to K562 cells membrane was observed by phase-contrast microscopy and flow cytometry. NK cells were purified and expanded from PBMC, MTT method was used to determine the effect of platelet on the lysis action of K562 by NK cells. Result: K562 cells can adhere to platelets in vitro. The adhesion ratio was 25.0±4.3%. Platelets can inhibit the cytotoxic action of NK cells against K562 cells in vitro. The degree of inhibition depends on the amount of platelets. Conclusion: Platelets can interact with K562 cells in vitro and inhibit the cytotoxic action of NK cells against K562 cells.
