摘要
本文以人外周血淋巴细胞为材料,用超抗原葡萄球菌肠毒素B(SEB)刺激后,检查了淋巴细胞的应答反应及特征。结果显示出微量的SEB(100ng)具有比PHA、SEA更为敏感的诱导T细胞应答反应的能力。T细胞应答反应中粘附性巨噬细胞起了抗原提呈作用;去除了粘附细胞,T细胞表现出完全不反应。T细胞活化途中伴随白介素2(IL-2)的释放。荧光染色的结果证明SEB诱导了外周血T细胞的增殖,这些细胞是识别MHCII类抗原的T细胞;它们由最初的27%增加到41%。并显示出由外周血群细胞分化而来。提示外周血细胞的大量增殖可能与外周血T细胞分化有关。
Our study focused on extrathymic differentiation of T cells.Staphylococcal enterotoxins B(SEB),as one kind of superantigen,was used and induced massive proliferation of human peripherialblood T cells.Adherent cells in lymphocytes,rather than non-adherent cells,also played an importantantigen presenting role during T cells proliferation,After SEB-stimulating,IL-2 were released andpopulation increase from 27%to 41%.They were differentiated from population. Theresults showed that superantigen SEB may up-regulate the response reaction of T cells in periphe-rial blood lymphocytes and the proliferation of Tcells may be involved to extrathymic differentia-tion of T cells.To study mechanism of T cells differentiation will give us new means to fight disease,and theknowledge about T-cell activation will help us to modulate the immune system in new ways.
