摘要
本研究参考公开发表的Alfort株基因组全序列 ,用计算机软件Dnastar的PrimerSelect程序设计了两对SCFVE2基因高变区的特异性引物 ,对黑龙江东部地区分离的 5株猪瘟野毒 (HL、JN、MSND、MSPD和MSHT株 )进行RT_PCR扩增、克隆和序列测定 ,并将这 5株野毒的高变区基因序列与国内外公开发表的参考毒株的相应部分进行了比较。结果表明 ,5株分离毒均为基因II群 ,且形成两个独立的分支 ,HL、JN株与HCLV株氨基酸差异在 60 %以上 ,而MS株与HCLV株氨基酸差异在 3 0 %左右。
Five strains of classical swine fever viruses,HL、JN、MSND、MSPD、MSHT,isolated from some areas of Heilongjiang province are investigated.According to the published gene sequence of the Alfort strain,two pairs of specific primers for amplifying various domain of E2 gene were designed through program PrimerSelect of Dnastar software and synthesized.A fragment of E2 gene of each strain was amplified by RT_PCR and cloned into T vector and sequenced.The amplified gene sequences of these five field strains were compared with that of published CSFV strains.The results show that the five strains belong to GroupII but two independent subgroups,and the difference between HCL V and HL,JN is more than 60 %,and the difference between HCL V and MS is 30 % or so.
