摘要
在中枢神经系统微血管形态学研究方面,首次应用钨酸钠(ST)作为稳定剂,以TMB—ST作为成色方法用于过氧化物酶组织化学法显示中枢神经系统微血管构筑。通过对过氧化物酶—TMB—ST法、碱性磷酸酶法(钙铅法)和腺苷三磷酸法8种组织化学法所显示的大鼠脑及脊髓微血管构型的比较观察,以及应用电子计算机图像分析处理系统和光镜下体视学测试系统对微血管的计量学分析比较,表明:过氧化物酶—TMB—ST 法是显示中枢神统微血管的良好方法,具有反应灵敏、产物稳定、方法简便等优点。
In the region of morphoe mtric investigating of miernvasculature ofthe central nervous system,we were the first to use sodium tungstate(st)as astabilizer of TMB in order to improve the quality of peroxidase histochemistrymethod to reveal microvasculature of the central nervous system.Through thecomparative observation and stereologic quantitative analysis of microvasculat-ure stained by per(?)xidase histochemistry(TMB—ST)method,alkaline phosphat-ase histochemistry method and adenosine tripnosphatase histochemistry method,we suggest that endogenous peroxidase activity of red cells is an excellent mar-ker for revealing the central nervous system blood vessels at the light microscopiclevel.The advantages of the new peroxidase histochemistry(TMB—ST)methodare sensitivity in reaction,stability of its product,and convenience in perform-ance.
