摘要
目的 探讨α 甘草酸对豚鼠肾脏 11β 羟基类固醇脱氢酶 (11β OHSD2 )的抑制作用。方法 运用微粒体酶分析技术 ,观察α 甘草酸减小微粒体酶对底物皮质醇的转化率 ,并测定酶的动力学常数。结果 α 甘草酸能够减小微粒体酶对底物皮质醇的转化率 ,和阴性对照、β 甘草酸相比 ,差异均有显著性意义 ;动力学常数Vmax=15 .2 5nmol/mg .h ,Km=1.31μmol/L ,Ki=(2 0 9.2± 17.1) μmol/L。 结论 α 甘草酸能够抑制豚鼠肾脏 11β OHSD2 ,但和 β 甘草酸相比 。
OBJECTIVE To demonstrate the inhibitory effects of α glycyrrhizic acid on 11β hydroxysteroid dehydrogenase from Hartley guinea pig kidney. METHOD Using microsomes from the kidney cortex,the reduction of transform rate of cortisol by α glycyrrhizic acid was observed and kinetic constants were determined.RESULTS Compared with negative group and β glycyrrhizic acid, α glycyrrhizic acid reduced transform rate of cortisol with a Ki of 209.2μmol/L±17.1μmol/L,V max =15.25 nmol/ mg.h,K m=1.31μmol/L.CONCLUSION Compared with β glycyrrhizic acid,α glycyrrhizic acid can inhibite the 11β hydroxysteroid dehydrogenase from Hartley guinea pig kidney,but was less potent than β glycyrrhizic acid.
