摘要
研究了一株产淀粉酶酵母-扣囊拟内孢霉的原生质体形式与再生条件,通过正交实验研究了菌龄、酶浓度、酶解时间、酶解温度、稳渗剂种类等对原生质体形成与再生的影响。结果表明,对数生长期早期的细胞(18~24h),用1.5%的蜗牛酶液,0.7mol/L的KCl做渗透压稳定剂,30℃水浴酶解60min,可获得最佳的形成率与再生率。
The suitable condition of protoplast formation and regeneration of Endomycopsis Fibnligera protoplast were studied. Through various experiments, we studied the influence of the age of the yeast, the potency of snailase enzyme, the enzymolysis time, temperature and the stablizers on the protoplast formation and regeneration.The results shows that it was beneficial under the conditions of using early exponential phase cell(18 ~ 24h), 1.5%snailase stablized with 0.7mol/L KCl and digesting for 60min at 30℃.
