抗纤软肝冲剂对肝星状细胞自分泌转化生长因子β_1的影响 被引量:2

Serapharmacological Effect of Kangxian Ruangan Granule on the Self-secretion and the Gene Expression of Transforming Growth Factor beta-1 of Hepatic Stellate Cells

摘要 目的:从细胞分子水平研究抗纤软肝冲剂对肝星状细胞(HSC)表达转化生长因子β_1(TGF-β_1)的影响。方法:抗纤软肝冲剂药物血清温育传一代HSC,以0.85%氯化钠溶液和秋水仙碱为对照。免疫细胞化学分析TGF-β_1的蛋白合成;半定量RT-PCR法检测TGF-β_1 mRNA的表达量。结果:抗纤软肝冲剂组TGF-β_1蛋白表达量为108.41±4.50,mRNA相对表达量为54.41±5.57,明显低于0.85%氯化钠溶液组和秋水仙碱组(P<0.01)。。结论:抗纤软肝冲剂能抑制HSC的TGF-β_1基因和蛋白表达,阻断其自分泌放大活化效应,这可能是该方抗肝纤维化的细胞分子学机制之一。 Objective: To study the effect of Kangxian Ruangan Granule (KRG) on the expression of TGF-β1 at the cellular and molecular lever. Methods: The medicated sera were prepared to incubate the subcultured HSC, and sodium chloride sera and colchicines sera were taken as control groups. TGF-β1 protein production was semiquantified by immunocytochemistry stain, gene expression of TGF-β1 was analyzed by semiquantitative RT-PCR. Results: KRG obviously inhibited the expression of TGF-β1 mRNA and its self-secretion. Conclusion: KRG could inhibit the gene expression and protein secretion of TGF-β1 of HSC, and obstruct its amplified auto-activation, which might be the cellular and molecular mechanism of its antifibrotic action.
