甲磺酸甲酯对CAOV3细胞端粒酶活性的影响

摘要 目的:探讨卵巢癌细胞CAOV3经烷化剂甲磺酸甲酯(MMS)作用后端粒酶活性变化与生长状态之间的关系。方法:台盼蓝拒染法计数活细胞,噻唑蓝(MTT)法测定药物作用浓度,流式细胞术观察细胞凋亡,端粒重复扩增法(TRAP)、聚丙烯酰胺凝胶电泳和银染法检测端粒酶活性。结果:用药组与未用药组活细胞百分率无显著差异。MMS主要使CAOV3细胞受阻于S期,未见凋亡峰。用药组与对照组相比较端粒酶活性增强。结论:MMS对CAOV3细胞端粒酶活性有上调作用。CAOVB细胞对MMS的抗凋亡效应可能与端粒酶活性上调有关。 Objective: Our aim was to investigate the relation between the growth and change of telomerase activity in ovarian cancer cell CAOV3 line after exposure to methylmethanesulfonate ( MMS ). Methods : Trypan blue exclusion was used to count the number of alive cells. Concentrations of MMS were determined by using MTT test. Apoptosis of the cells was observed with flow cytometric analysis. The activity of telomerase was examined by TRAP, polyacrylamide gel electro-phoresis, and silver staining. Results: The difference of the percentages of viable cells among MMS groups and control group was not obvious. The cells were mainly arrested at S phase after MMS treatment. No sub-G, fraction was observed. Compared with control group,the telomerase activity of the cells increased after treatment with MMS. Conclusion:The telomerase activity of CAOV3 cell can be up-regulated by MMS. The anti-apoptosis of CAOV3 cell to MMS may be related with the up-regulation of telomerase activity.
