羊膜及培养鸡角膜缘上皮对外周血T细胞CD 69分子的活化研究 被引量:3

摘要 目的 研究不同状态的羊膜、鸡角膜浅板层和培养鸡角膜缘上皮细胞,对人外周血T细胞的刺激.以及转化生长因子-β1(TGF-β1)对T细胞表面活化分子CD 69表达的影响。 方法 随机采取6例健康成人外周血,制备不同状态的羊膜、鸡角膜缘上皮细胞和鸡角膜浅板层并联合羊膜作刺激原,设立阴性对照组及阳性对照组刺激原.将10组刺激原分别与全血混合培养36 h,标记检测的抗体,用流式细胞仪分析。 结果 未见新鲜羊膜、去上皮新鲜羊膜及保存羊膜对外周血 T细胞及CD 8+T细胞表面分子CD 69产生活化表达;以羊膜为底物培养鸡角膜缘上皮细胞、鸡角膜浅板层未使人外周血 T细胞及CD 8+T细咆亚群表面分子CD 69产生活化表达;TGF-β1可明显下调人外周血T细胞及CD8+T细胞亚群表面分子CD69的活化表达。 结论 进一步支持羊膜免疫原性很低的观点;单用或联合羊膜应用培养鸡角膜缘上皮细胞和鸦角膜浅板层对人外周血T细胞及CD 8+T细胞亚群免疫刺激作用很弱;进一步支持TGF-β1有明显免疫抑制作用的观点。 Objective To study the stimulation of amniotic membrane, chicken lamellar cornea and the cultured chicken limbal epithelial cells to T cells in human peripheral blood in vitro and the effect of transforming growth factor-β1 (TGF-bbbbbbb1) on the expression of CD 69 molecule in T lymphocyte surface. Methods Six healthy adults were selected , each person offered peripheral blood 1 ml. Amniotic membrane, chicken limbal epithelial cells and chicken shallow lamella cornea were prepared and the amniotic membrane was used as stimulative antigen. After 36-hour incubation, the cell was labeled by fluorescence monoclonal antibody and analyzed by cytometry for the expression of CD69. Results The expression of CD 69 was negative on the T lymphocyte in peripheral blood,CD8 + T lymphocyte stimulated by fresh aninion , fresh amniotic membrane without epithelium and preserved amniotic membrane. There were no active expression of CD 69 on the peripheral T lymphocyte, CD 8+ T lymphocyte stimulated by the chicken shallow lamellar cornea,the chicken limbal epithelial cells cultured with amniotic membrane. TGF-β1 down-regulated the activating expression ol CD 69 on the T lymphocyte of human peripheral blood and CD 8 + T lymphoeyte subset. Conclusions It is suggested that the immugen in amniotic membrane is low. The stimulation to T lymphocyte of human peripheral blood and CD8+ T lymphocyte subset is very weak with cultured chicken hmbal epithelial cell and chicken lamellar cornea or combined with amniotic membrane. TGF-β1 shows obvious immune inhibition.
