摘要
采用反相高效液相色谱法 ,在C1 8柱上以甲醇 水为流动相进行梯度洗脱 ,在 0~ 1 0min内将葛根黄酮提取物进行分离 ,并在 2 5 0nm检测 ,采用标准加入法对实际样品中的葛根素含量进行了分析。结果表明 :平均回收率为 94 .8% ,RSD为 2 .4 %。
A method for the separation and determination of Pueraria lobata isoflavonoid extract by reversed-phase high performance liquid chromatography (RP-HPLC) was proposed. All the components can be separated through a C-18 analytical column with methanol-water as mobile phase and a gradient elution-changing the percentage of the methanol from 15% to 58% in 10 minutes. Puerarin were quantitated by standard addition method at the detection wavelength of 250 nm. The result shows that the average recovery of Pueranin is 94.8% RSD is 2.4%.
