酸性成纤维细胞生长因子修复脑血管病后神经细胞损伤机制 被引量:3

摘要 目的:探讨酸性成纤维细胞因子(aFGF)对地塞米松诱导的肾上腺髓质嗜铬细胞瘤PC12细胞凋亡的保护作用及其临床意义。方法:采取体外细胞培养PC12细胞的方法,制备地塞米松诱导的细胞凋亡模型,再使用酸性成纤维细胞生长因子加入培养细胞,通过流式细胞仪观察凋亡细胞数量的变化情况。结果:地塞米松可以诱导肾上腺髓质嗜铬细胞瘤PC12细胞株发生凋亡,发生的比例是18.59%±0.38%,当使用aFGF处理损伤的细胞后,凋亡的发生比例明显下降,仅为10.05%±0.23%。结论:酸性成纤维细胞生长因子可以保护地塞米松诱导的肾上腺髓质嗜铬细胞瘤PC12细胞株,使其避免凋亡。aFGF可能具有保护缺血性脑血管病的神经细胞损伤的作用。 AIM:To investigate the protective effect of acid fibroblast growth factor on the apoptotic pheochromocytoma PC12 cells by aeroseb D inducing, and explore the clinical significance.METHODS:Using PC12 cells as the damaged model of aeroseb D inducing neurons, the apoptosis ratio by aeroseb D induced and the protective effect of rhaFGF was observed with flow cytometry technique.RESULTS:Treatment of PC12 cells with aeroseb D for 30 min, results revealed that the apoptosis ratio was 18.59%±0.38%,more higher than that of control 10.04%±0.23%(P< 0.01).CONCLUSION:Induction of apoptosis by aeroseb D can be inhibited by aFGF. aFGF can avoid lesion of neurocyte due to ischemic cerebrovascular disease.
