摘要
目的研究人脑挫裂伤后神经细胞的凋亡及凋亡相关基因Bcl-2、Bax在蛋白水平的表达。方法采用末端脱氧核苷酸转移酶介导的生物素脱氧尿嘧啶核苷酸缺口末端标记法(TUNEL),免疫组织化学方法检测人脑挫裂伤后急诊行开颅脑挫裂失活组织清除术和/或额极、颞极切除内减压术所得脑组织神经细胞的凋亡和Bcl-2、Bax蛋白的表达。结果15例研究病人中所有病人标本都表达Bax蛋白,8例表达Bcl-2蛋白,10例表达TUNEL阳性染色细胞。表达Bcl-2蛋白患者比不表达Bcl-2蛋白患者生存率明显升高(P=0.01)。结论虽然人脑挫裂伤的主要病理改变是坏死,但实验表明人脑挫裂伤后存在神经细胞的凋亡现象,Bcl-2基因家族参与了人脑挫裂伤后神经细胞凋亡。脑挫裂伤患者表达Bcl-2蛋白与预后有一定关系。
Objective To explore whether there was evidence of cellular apoptosis in clinically collected materials from human head trauma patients,as well as to attempt to determine the pathway by which it may occur.Methods Terminal deoxynucleotide transferase-mediated nick end labeling(TUNEL) were employed to detect the presence cell.Immunohistochemistry were employed to detect possible expression of the apoptosis related gene Bcl 2、Bax.Results TUNEL Postive cells were noted in ten patients,Bax expression was detected in all 15 patients,Bcl 2 expression in only 8 patients.Patients with Bcl 2 expression had a better survival rate than patients in whom no Bcl 2 expression was noted (P=0.01).Conclusion The results indicate that apoptosis does occur in patients after traumatic brain injure.patients in whom anti apoptotic Bcl 2 is induced seem to have a better prognosis.
