摘要 目的探讨谷氨酰胺对肝脏移植后应激蛋白表达的影响。方法A组10例为正常肝脏;B组10例肝脏移植后不用丙氨酰谷氨酰胺二肽(Ala-Gln);C组8例肝脏移植后用Ala-Gln。观察标本的病理学改变和患者的营养状况,用免疫组织化学及原位杂交方法检测HSP70、HO-1蛋白和mRNA表达水平。结果B组和C组均未见免疫排斥反应,营养状况尚可;HSP70、HO-1蛋白及mRNA表达水平移植肝脏均升高,且应用Ala-Gln后更高。结论谷氨酰胺能增加供肝细胞应激蛋白基因转录和表达,加强肝细胞自我保护作用。 Objective To explore the impact of glutamine on stress protein expression af ter liver transp-lantation.Methods Classified10normal liver sample and18pu nctured donor liver sample as A(control)group,B(no Ala-Gln)group,C(Ala-Gl n)group according to the administration of Ala-Gln.Pathology and nutritional s tatus of all of patient were observed as well as the expression of protein and mRNA of HSP70and HO-1were performed immunohistochemistry analysis and in situ hybridizition.Results No rejection was found in B and C group.The nutritiona l status was good in all transplanted patients.The expression of protein and mR NA of HSP70and HO-1was increased after transplantation and was higher when ad ministated Ala-Gln.Conclusion Glutamine can increase the gene transcription and protein expression of the stress protein in hepatocyte after liver transplantat ion.That can augment self-protection of hepatocyte.
