摘要
目的 :观察甘氨酸 -钼磷酸盐 [( Gly H) 4 PMo12 O40 · 5 H2 O,GP]对肿瘤细胞生长的抑制作用及对荷瘤小鼠的抑瘤效应。方法 :采用 MTT法进行体外抑瘤效应测定 ,建立小鼠荷 S1 80腹水瘤模型 ,测定体重增长率、抑瘤率以及延命率。结果 :体外实验表明 ,实验浓度的 GP对正常组织细胞均无细胞毒性作用 ( P<0 .0 5 )。对宫颈癌 Hela细胞株、前列腺癌 PC- 3m细胞株、小鼠腹水瘤 S1 80细胞株、原代肺癌细胞等肿瘤细胞生长有明显抑制作用 ( P<0 .0 5 ) ,并呈一定的量效关系。其中对人前列腺癌 PC- 3m细胞株尤为明显。体内实验表明 ,给药组能够抑制 S1 80腹水瘤生长 ,明显延长小鼠生命期 ,延命率高于 CYT阳性对照组。急性毒性实验 LD50 为 2 735 .2 7mg·kg-1,LD50 95 %可信限为 :1 931 .97~ 3872 .5 8mg· kg-1。结论 :本研究表明 ,GP能抑制多种人癌细胞生长 ,延长荷瘤小鼠生存期 ;GP属低毒。
Objective:To study the anti tumor activity and toxicity of GP Methods:MTT method was used to examine the cytotoxicity of GP in normal cells and the cytotoxic activity of GP against several human cancer cell lines S180 tumor bearing mice used as an animal model,the tumor inhibition, survival days and acute toxicity of GP were measured Result:The results showed that there were no toxic activities in the range of the experimental concentration, and GP could inhibit the growth and proliferation of human carcinoma cells listed above in a dose dependent manner in vitro. Furthermore, it was able to kill PC 3m cell significantly and thus showed the potential value in the treatment of human prostate cancer. The results also revealed that increase of tumor weights in mouse was obviously inhibited, and death of the tumor bearing mice was delayed and the survival rate was raised. The median lethal dose (LD 50 ) of GP in mice was 2 735.27 mg·kg -1 , and the ranges of 95% confidence limit was 1 931.97~ 3 872.58 mg·kg -1 . It indicated that GP was a low toxic material. Conclusion:As a low toxic material,it is found that GP have potential application in anti tumor therapy
