摘要
目的 :建立一种快速、稳定、灵敏并可反映细菌活性的细菌计数方法。方法 :以大肠杆菌为研究对象 ,探讨MTT法用于细菌计数的可行性及反应时间、测量细菌数量的范围、MTT的用量、是否加入溶解剂等实验条件。结果 :细菌数在 3.8× 10 7~ 1.2× 10 9 ml范围内测出的OD值与细菌浓度呈良好的正相关 ,反应时间为 2 0分钟 ,MTT10 μl、2 0 μl均可 ,2 0 μl效果更佳。可不用溶解剂。 结论
Objective:To establish a rapid,steady and sensitive germ counting method that could reflect the germ's activity. Methods:E.coil was employed to determine the conditions of assay,which include the feasibility,reaction periods,germ's number concentration of MTT and so on.Results:The OD has a direct relativity with the concentration of germ when the number of the germ is in the period of 3.8×10 7~1.2×10 9/ml by reacting with 10 or 20μl MTT,(20μl is better),without dissolvent matter.Conclusion:MTT colorimetry could be used to measure the amount of live germ.
