摘要
以拮抗菌——地衣芽孢杆菌为研究对象,根据其对其它常见病原细菌具有抑制性的特点,利用吖啶橙的诱变作用,使其发生突变,并通过单菌落琼脂块抑菌圈法筛选获得高抑菌性的突变菌株;采用滤纸片抑菌环法、MTT-四氯化碳萃取吸光光度法、平板计数法和分光光度法等对其抑菌性以及抗菌谱进行测定,确定所得突变菌株为高稳定性、高抑菌性和抗菌谱较广的地衣芽孢杆菌,并考虑将此菌株应用于医药行业或基因工程的目的菌株。
This study took the antagonistic bacterium as the research object. Based on the character of resisting common pathogen of this bacterium, and taking advantage of the mutagenesis effect of acredine orange, the antagonistic bacterium was induced to mutate. Then, the mutant bactrium strain of high antifungal activity was obtained by screening the mutant colony borrowing the piece of angragar inhibition zone method of single bacte-rium colony. Lastly, the genetic stability, efficiency value of the bacterium resistance and the span of the bacte-rium it resisted were measured by filter paper inhibition zone method, MTT-CCl4 extraction method, slab counting method and turbidimetry method, and it was comfirmed as highly stable, highly antibacterial and broad in antibacterial span, and it could be considered to be untilized in pharmaceutical or in gene engineering field as target bacterium.
