食管癌中DNA聚合酶β的突变 被引量:2

摘要 目的 研究DNA聚合酶 β基因在采自食管癌高发区的食管癌标本中的突变情况。 方法 提取总RNA反转录合成cDNA ,经PCR扩增后以SSCP检测其变异情况。结果 在食管癌组织标本中存在polβ基因的突变 :2 0例癌组织标本中发现 9例突变 (45 %、9/ 2 0 ) ,癌旁组织中发现 1例突变 (5 %、1 / 2 0 )。结论 在食管癌组织标本中确实存在polβ基因的突变 ,推测在食管癌的的发生发展过程中polβ基因突变是一个始动因素 ,是各种致癌因子作用的枢纽 ,由于该基因的突变可能使得癌基因、抑癌基因如 :ras、p53、Rb等的原发性损伤得不到及时正确修复 ,导致细胞周期的失控、增殖过多 。 Objective To detect the DNA pol β gene mutation in esophageal carcinoma.Methods Extracting total RNA from esophageal carcinoma,corresponding cancer specimens tissues and normal tissues,synthesizing cDNA and then using them as templates proceed PCR,the products of PCR were checked by SSCP.Results There were 9 samples detected mutation in 20esophegeal carcinoma,1 sample detected mutation in 20 corresponding cancer speciment tissues and no mutation in normal tissues.Conclusions There were pol β gene mutations in esophageal carcinoma.That suspected pol β gene mutations were the initiation factors in the process of esophageal carcinomagensis.Mutations of pol β gene may cause the primarty impairments been not repaired,and may lead to losing conduction in cell cycle.Then the cell may proliferate excessive and may accelerate carcinomagensis. [
