IL-18在系统性红斑狼疮外周血单个核细胞中的表达

摘要 目的:探讨系统性红斑狼疮(SLE)IL-18的表达及其和疾病活动的关系。方法:以β-actin为内参照,用半定量逆转录多聚酶链反应(RT-PCR)法测定16例SLE患者及15例正常对照外周血单个核细胞(PBMC)中IL-18mRNA水平,观察IL-18与疾病活动指标狼疮活动指数(SLEDAI)、血沉(ESR)的相关性。结累:SLE患者IL-18mRNA表达与正常组相比增高非常显著(1.526±0.437vs0.453±0.290,P<0.001)。相关分析显示:SLE组IL-18mRNA与SLEDAI、ESR显著正相关(n=16,r分别为:0.649、0.573。P分别小于0.01、0.05)。结论:SLE患者IL-18mRNA表达水平显著高于正常对照组,且与SLE患者疾病活动性相关。 Object:To further investigate the expression of IL-18 in patients with systemic lupus erythematosus (SLE), and the relationship exists between the expression of IL-18 and activities of the disease. Method: expression of mRNA in peripheral blood mononudear cells (PBMC) from in 16 patients with SLE and 15 healthy subjects were detected by semi - quantitive RT-PCR. Results: 1). PBMC from patients with SLE were found to contain greater levels of IL18 mRNA than that from healthy subjects (1.526+0. 437 vs 0. 453 + 0. 290, P< 0. 001); 2 ). IL-18 mRNA levels were correlated to the disease activities as assessed by levels of SLE disease activity index (SLEDAI), erythrocyte sedimentation rate(ESR)( n=16, r=0.649 and 0.573, P less than 0.01 and 0.05, respectively). Conclusion: IL-18 is among a matrix of inflammatory cytokines produced abundantly in patients with SLE and is associated with activities of the disease.
