α平滑肌肌动蛋白和转化生长因子β_1在小管间质纤维化模型大鼠肾组织中的表达 被引量:8

摘要 目的 探讨肾小管上皮细胞 -肌纤维母细胞表型转化在肾间质纤维化形成中的作用。 方法 制备大鼠单侧输尿管梗阻模型 ,在光学显微镜下观察大鼠肾组织病理变化 ,采用免疫组化法检测肾组织中α平滑肌肌动蛋白 (α -SMA)和转化生长因子β1(TGF- β1)的表达 ,并采用计算机图像软件进行定量分析。结果 肾脏病理检查显示形态学改变符合肾小管间质纤维化的病理变化 ;在输尿管梗阻14、28d后肾组织α -SMA和TGF - β1表达较对照组增加 (P<0.05)。 结论 α -SMA可作为肾小管上皮细胞 -肌纤维母细胞表型转化的重要标志物 ,TGF- β1 可能参与了肾小管上皮细胞 -肌纤维母细胞表型转化 ,且是进一步导致肾间质纤维化的重要因素。 Objective To study the role of tubular epithelial-myofibroblast transdifferentiation in pathogenesis of renal interstitial fibrosis.Methods Immunohistochemistry staining was used to investigate the pathological change of kidney and the expression of α-smooth muscular actin(α-SMA) and transforming growth factor β1(TGF-β1) in renal tissue of rats with unilateral urethral obstruction (UUO).Results The levels of expression of α-SMA and TGF-β1 on the 14th day and 28th day in kidney of UUO were significantly higher than that of normal rats(P<0.05).Conclusion α-SMA may be used as a marker of the tubular epithelial-myofibroblast transdifferentitiation;TGF-β1 may play an important role in pathogenesis of renal interstitial fibrosis.
