摘要 目的 研究VEGF反义寡脱氧核苷酸(AODN)对大鼠C6胶质瘤生长的影响,探讨抗血管生成作为胶质瘤辅助治疗新途径的可能性。方法 制备大鼠皮下C6胶质瘤模型,在接种细胞后不同时间点局部给予VEGF-AODN和其他处理因素,观察皮下肿瘤形成情况,测量肿瘤体积及重量,TUNEL法和电镜技术检测瘤细胞凋亡。结果 接种72h后开始局部注射反义寡核苷酸,使大鼠皮下C6胶质瘤的成瘤潜伏期延长,为(25.13±3.00)d,肿瘤重量减轻,为(1.13±0.17)g;皮下局部肿瘤形成后连续注射AODN,使肿瘤体积缩小,瘤细胞凋亡增加,瘤内坏死区扩大,但对细胞增殖无明显影响;电镜观察到AODN治疗组微血管基膜增厚,管径变小。结论 抗血管生成治疗可望成为胶质瘤治疗的一种辅助手段,具有临床应用前景。 Objective To observe the inhibitory effect of VEGF antisense oligodeoxynucleotides (AODN) on C6 glioma growth in rats, and explore the probability of antiangiogenesis as a new therapeutic strategy for glioma. Methods C6 glioma cells were subcuta-neously injected in rats. Then VEGF AODN and control drugs were intratumoral given respectively. The volume and weight of subcutaneous tumor were measured. Tumor cells apoptosis and the vascular structure were detected by TUNEL method and electron microscope technique. Results There are longer latent of the tumor formation and lighter weight of the tumor in the rats treated by AODN as compared with the control rats. Treatment with AODN also induced the apoptosis of the tumor cells and had no effect on the tumor cells proliferation. Electronscope observation showed that vessel diameter became smaller and basic membrane became thicker in AODN group than those in the control group. Conclusions The antiangiogenesis therapy may have clinical utility as a new therapeutical method for the gliomas.
