摘要
目的分析胶质瘤中信号传导和转录活化因子3(STAT 3)和血管内皮生长因子(VEGF)的表达与脑胶质瘤恶性程度的关系。方法采用免疫组织化学方法检测70例人脑胶质瘤和11例正常脑组织中STAT 3和VEGF的表达。结果胶质瘤中STAT 3和VEGF的表达水平在高级别(Ⅲ,Ⅳ级)组明显高于低级别(Ⅰ,Ⅱ级)组,两组间有显著性差异(P<0.01),STAT 3的表达和VEGF的表达呈显著正相关(P<0.01)。结论STAT 3和VEGF的表达与胶质瘤的恶性程度有密切关系,且两者协同在胶质瘤发生、发展过程中起重要作用。两者的相关性则提示VEGF基因由STAT 3蛋白调节。抑制STAT 3蛋白通路可能有望成为一条治疗胶质瘤的新途径。
Objective To explore the relationship between the expressions of signal transducer and activator of transcription 3 (STAT 3) and vascular endothelial growth factor (VEGF) in human gliomas and the degree of malignance, and their significance, Methods The expressions of STAT 3 and VEGF in 70 cases of glioma and 11 cases of normal brain tissues were detected by immunohistochemical assay. Results The positive rates of expressions of STAT 3 and VEGF were significantly higher (P〈0.01) in the gliomas of high grade (Ⅲ, Ⅳ) than that in the gliomas of low grade (Ⅰ,Ⅱ ). The expression of STAT 3 was positively correlated to the expression of VEGF (P〈0.01) in the glioma. Conclusions The expressions of STAT 3 and VEGF are close related to the degree of ,glioma malignance, and they play an important role together in the pathogenesis and development of glioma. It is suggested that VEGF gene is regulated directly by STAT 3 protein. The inhibition of STAT 3 pathway may become a new way to treat glioma.
